چکیده

Objectives: Naturally occurring regulatory T cells (nTregs) play an important role in immunologic tolerance and control immune-mediated pathology in murine models of autoimmune hemolytic anemia. Our aim was to measure nTregs and levels of interleukin (IL) 10 and IL-12 in peripheral blood mononuclear cell (PBMC) cultures from patients with idiopathic warm autoimmune hemolytic anemia (wAIHA) in an attempt to unravel some of the mysteries behind the pathogenesis of this autoimmune disorder. Methods: Twenty-seven patients with idiopathic wAIHA and 15 ageand sex-matched controls underwent fl ow cytometric analysis of CD4+CD25highFoxP3+T cells (nTregs) and analysis by enzyme-linked immunosorbent assay of IL-10 and IL-12 in the supernatants of basal and lipopolysaccharide (LPS)-stimulated PBMC cultures. Results: The mean (SD) percentage of circulating CD4+ nTregs in peripheral blood was signifi cantly lower in patients (4.63% [1.0%]) than in controls (9.76% [0.78%]) (P<.001). PBMCs from patients had signifi cantly higher basal levels of IL-10 and IL-12, with a dramatic reduction in responsiveness to LPS in vitro compared to controls. There was a signifi cantly negative correlation between the percentage of nTregs and reticulocyte count (RC), basal IL-10, and LPS-stimulated IL-10, and a signifi cantly positive correlation with haptoglobin (Hp) (P<.05). Basal IL-10 and LPS-stimulated IL-10 were positively correlated with RC (P<.001 in both cases) and negatively correlated with Hp (P<.01 and P<.05, respectively). Conclusion: Our study indicates that a reduced percentage of nTregs and IL-10/IL-12 imbalance may play an essential role in the onset and/or maintenance of this AIHA.

Concurrent Atypical Hemolytic Uremic Syndrome and Autoimmune Hemolytic Anemia: a case report

Background: Atypical hemolytic uremic syndrome (aHUS) is a life-threatening and scarce disorder characterized by acute renal failure and disease, non-immune microangiopathic hemolytic anemia and thrombocytopenia, leading to end-stage renal failure or death, and consequently maybe accompanying by extra renal manifestations. Case report: We reported aHUS accompanied by autoimmune hemolytic anemi...

Autoimmune Hemolytic Anemia preceding the Diagnosis of Primary Central Nervous System Lymphoma

In this study, a 2.5-year-old boy suffering from a febrile seizure with normal laboratory tests and a history of immune hemolytic anemia was examined. Brain MRI demonstrated some tumors in the frontal, parietal, and occipital lobe that corroborated the pathology results of primary central nervous system lymphoma for the patient. The patient was treated with high- dose of Methotrexate. Our re...

Autoimmune Hemolytic Anemia in a Patient with Probable Ataxia Telangiectasia: A Case Report

Background: Ataxia telangiectasia (AT) is one of the combined immunodeficiency syndromes with immunologic, neurologic, endocrinologic, hepatic and cutaneous abnormalities. Regarding the fact that autoimmune disorders; such as autoimmune hemolytic anemia (AIHA), are not generally expected in the course of AT, we present a patient with an unusual presentation of these two conditions. Case present...

A Fetal Hemolytic Anemia in a Child with Cytomegalovirus Infection

Background Autoimmune hemolytic anemia is a hematologic disorder that is rarely observed in infants and young children. Most of the cases are associated with viral or bacterial infections. In some cases, AIHA can be characterized by a chronic course and an unsatisfactory control of hemolysis, thus requiring prolonged immunosuppressive therapy. Case report Especially in children younger...

H2 control of natural T regulatory cell frequency in the lymph node correlates with susceptibility to day 3 thymectomy-induced autoimmune disease.

Day 3 thymectomy (D3Tx) results in a loss of peripheral tolerance mediated by natural regulatory T cells (nTregs) and development of autoimmune ovarian dysgenesis (AOD) and autoimmune dacryoadenitis (ADA) in A/J and (C57BL/6J × A/J) F(1) hybrids (B6A), but not in C57BL/6J (B6) mice. Previously, using quantitative trait locus (QTL) linkage analysis, we showed that D3Tx-AOD is controlled by five ...
